University of North Texas - BIOL 3520Lab report 2>Species hierarchy based on proteomics

Document Content and Description Below

Abstract With new research into the proteomic field has revealed many modification systems of one gene to code for many proteins and mechanisms that regulate the locations and expression levels of p... roteins. Comparing the morphology of organisms, including sizes, shapes, and developmental structures of both living organisms and fossils is the traditional way evolutionary trees were constructed. This will be used to construct a cladogram of 5 tissues samples from unknow origins by protein molecular weights of the muscle proteins: Actin and Myosin light chain. By using SDS-PAGE (Sodium dodecyl sulfate – polyacrylamide gel electrophoresis) a form of electrophoresis that treats samples with SDS to denature proteins. Since SDS-PAGE separates according to their mobility, the samples having identical charge to mass ratios results in fractionation by size. Then using the Western Blot as an analytical technique used to detect specific proteins in each sample of tissue homogenate or extract. Which will help confirm the myosin proteins in muscle tissue for the sample order in the cladogram. The results support that the idea of using proteins as a classification of hierarchy challenges to old methods phylogeny and can pave the way to for the ability for the scientific community to accurately construct future evolutionary trees [Show More]
