Explain how you would use a mutagenesis screen to identify cell division cycle mutants in Schizosaccharomyces pombe. (5 marks) You would screen for conditional or temperature sensitive mutants because you would expect mutations in cell division cycle genes to be lethal (1 mark). The yeast is mutagenised in permissive conditions, where you would expect conditional mutants to have a wild-type phenotype (1 mark). The mutagenised yeast would be transferred to restrictive conditions to reveal a cell division cycle mutant phenotype (1 mark). Cell division cycle mutants do not progress through the cell cycle and arrest at a specific point of the cell cycle (1 mark). This distinguishes them from non-cell division cycle mutants, which would arrest immediately on transfer to the restrictive conditions irrespective of their progression through the cell cycle (1 mark).
