Students should work individually to answer the following questions and in creating the figures. Week 2: Questions 12 - 15 12. Why does DNA move through an agarose gel? The DNA is put into a gel that is in a field with an electrical current. The DNA is placed in wells on the negative side of the electrical current and become negatively charged. The negatively charged DNA molecules migrate towards the positive end of the field of agar gel immersed in agar bath. 13. What is an Allele Ladder? What is its function in DNA profiling? This is also known as a molecular weight ruler a sample of DNA composed of fragments that represent alleles that have similar fragments to the DNA you are testing. Each allele is a different size and therefore, there is a clear separation of the different alleles in the form of bands. The ladder’s function is to be used as a reference to compare and determine the product or suspects DNA band sizes so that matches can be determined. 14.
