Less wasteful, excess can be toxic/degrading  fusion proteins o yeast/phage/bacterial surface display – can display target protein on surface o take a given tumor -> grow in organism of choice, generate a polypeptide library o -> display on surface on yeast/bacteria -> insert into mice o -> some of the peptides stick to tumor region in mouse -> collect the tumor o After identifying the sequence:  Can tag again -> inject again -> stick to tumor again  Can be used in target therapy o CAR – chimeric antigen receptor  Create a synthetic fusion protein to be inserted into a T cell  Attach a signaling domain that activates the T cell -> attached antibody will recognize the antigen on the tumor cell  Otherwise, T cell would be unable to detect the presence of the tumor (use antigens that T cells don’t recognize as being dangerous)  Things to be careful with: can’t use T cells from another person  Will be recognized as foreign -> attacked  Have to use own T cells to prevent this  Alternatively: knock off/out the TCR receptors to prevent such an attack (using something like CRIPSR), allows one to use T cells from someone else in this way [Show More]
